Overtime mode gives you 24 hours (2 hours real time) to create a cure for the Zombie virus. Isabela tells you what to find and you have to run around the mall finding the stuff you need to cure or atleast slow the "Zombification" process. It's kinda' crappy if you ask me.

You can just kill non-stop without any missions in Infinity Mode. But your health gradually drains so you have to keep finding food to stay alive. And all you have to do is see how many days you can stay alive.
To get Infinity Mode, you have to get the best ending in 72 hour mode and beat Overtime Mode.
