Hiya. I found it took me forever to find where things were on this one. Once you have the big key and you have all the other keys (explore, explore, explore), go to the room with the big chest. Use the hook shot to move to the platform directly behind the chest (so, it would be at the top of your screen). You will be pulled by the hookshot and the skulls on the floor to that platform.
Go through the door and then you will enter a hallway with water, go to the right and you wil enter a whole room filled with water. The middle square stone can be pushed and I think you get coins or something. Anyway, go to the second water fall on the wall, second from the right of your screen, and go through it. You will find the Blob thing there. Use the hookshot to pull away the balloons and once you have the eye alone, keep moving, time the move it does, and hit it with your sword. I found it was the easiest one to defeat so far.
