It would be great to get stats upgrades and deadeye upgrade to 5.

Could someone explain to me how to run the game in Offline Mode so that I'm truly offline while playing and be able to use this trainer ?

You don't need to be offline to use the trainer, only in Single Player. Not Online or you risk getting banned. Single Player and Online are separate game modes. If you really need to play the game offline, you will need to generate an offline key.

Just a question. The trainer loads properly and states that it's working etc...but the stamina and health decreases...is that normal?

Not using unlimited health but noticed stamina decreasing when activated.

The same way you start every other single player game. Start the game through the launcher and then follow the on screen prompts and menus for single player.
I play all my games totally offline single player with internet disabled.
RDR2 PC is the first game I've bought that has no indication as to Single Player.
When I load the game all I get is 'Q Story/Online E ' at top right and at the bottom part of the screen there is also an option for 'Story ENTER '. Which one is Single Player ?
