You will have to do a bit of exploring for that. Missions that are located in unexplored areas are marked with a violet colored square with a question mark in it. Travel there and get that mission unveiled.

If you're talking about the Secret mission "Revelation" I think you need to go to the Police HQ and walk into 3 markers, then walk into an office to pick up something. Should make you go to the church to do a LOT of killing.
