wonder how we can get the characters to use in free play (im sure some wiill be unlocked in story mode) but as for the others if it will be like Pirates where we wander around the hub and purchase them or like Harry potter where we had to collect character tokens within the game itself and purchase them at a store in the main hub
If by "use" you mean switching to others, I'm unsure for this game *my copy will be here tomorrow*, but I know the ones I have, you hold the you use to switch between the characters in story mode, so for example, with an Xbox 360 controller, you hold Y

It's a bit like Lego PotC, the main heros (like Sups, GL, WW) you unlock during story mode, the villians & henchmen you have to find on the map and beat up then buy, & the other heros (Batgirl, Hawkman etc) and extra characters you buy from gold brick doors once you have the required number of bricks
run into the riddler thug outside arkham after the mission, had to beat up a few of them then one just stood there and had an action available, used it to buy him.
It is just like Pirates of the Caribbean, as darth_biggious said. Switching is also like PotC, even in free play mode (which is awesome!), but to switch when you are roaming the city, you need to beat all Spoiler:

If you want to get Gold Bricks more quickly, I would recommend flying around for a while with Superman getting the Gold Bricks that are in gold cages or behind the gold covers on the heads of the giant Statues all over as soon as you get him, before *BIG SPOILER* the kryptonite makes him unable to fly
