What I did was pop a crossbow bolt into his b**** ass to get his attention so he didn't run away, then called in a couple assassin waves and they got him off his horse, then I kicked him in the nuts a bunch of times and stuck my Spadone into his skull.

You can kill him in two ways

When he tries to kill you with a horse
Counter his attacks { Ezio will hit the horse Which allow the Borgia captain to fall down. Now kill Him }

Or Take a horse With hidden blade
lock him and air assassinate him

i often just walk around restricted area and kill off the guards first and hide around a corner and run up on the horse and take him off and kill him.

You can assassinate him via horse. Ride in, air assassinate him. However if you got a crossbow, you can shoot him once than duck into a building. Wait unti'll the guards lower suspicion than shoot him again. That worked for me too. There is a special place that is in the middle. You have to run there, it's a ruined building with a destroyed wall. You can shoot him from there and just duck behind the wall. I've never had issues with that spot.

It's near impossible though to do this without ducking after you shoot him. Guards will notice you before you can reload and the dude will run off or you can just arrow storm him.

i called in my assassins as the guards were distacted i ran up to the captain and pull him off the horse and use the blade.Also i hired mercinces as well which helped.
